When a child has SCD, the red blood cells are hard and sticky. They are shaped like the letter C (and like a farm tool called a sickle).

When they lose their oxygen, they form into the shape of a sickle or crescent, like the letter C.

What is a sickle shaped red blood cell?In sickle cell disease, the red blood cells contain an abnormal form of hemoglobin (the protein that carries oxygen). The abnormal form of hemoglobin is called hemoglobin S. When red blood cells contain hemoglobin S, they can become deformed into a sickle shape and less flexible. Not every red blood cell is sickle-shaped.

Sickle cell disease is an autosomal recessive disease, which means both parents should carry the abnormal gene for the child to have sickle cell disease. When the child inherits abnormal genes from only one parent, it is called a sickle cell trait. In sickle cell disease, the structure of hemoglobin which carries oxygen in our body is abnormal. So, when the oxygen demand is increased the red blood cell contorts forming a sickle cell, which gets destroyed owing to their abnormal shape causing anemia, and severe pain when these abnormal blood cells get clumped in blood vessels. In those with sickle cell trait, only mild symptoms are present, but they can pass on the disease to their children.

How does a sickle phenotype work?It does this by reversibly binding with the N-terminal valine of alpha (α) chain of Haemoglobin, changing its conformational structure and stabilizing the oxygenation form of the molecule. This reduces the concentration of deoxygenated HbS, which is the form of the molecule that polymerises to give the sickle phenotype.
What is Sickle Cell Anemia (SCD)?Sickle cell anemia, or sickle cell disease (SCD), is a genetic disease of the red blood cells (RBCs). Normally, RBCs are shaped like discs, which gives them the flexibility to travel through even the smallest blood vessels. However, with this disease, the RBCs have an abnormal crescent shape resembling a sickle.

Sickle cell anemia is caused due to point mutation. The GLU of 6th codon in beta globin chain is replaced by VAL. This type of mutant haemoglobin cause a change in RBC shape from biconcave to sickle shape, thereby leading to reduced oxygen carrying capacity.
